HART, LARRY G., ANTHONY M. GUARINO, AND RICHARD H, ADAMSON. Effects of phenobarbital on biliary excretion of organic acids 2*lz male and female rats. Am. J. Physiol. 2 17( 1) : 46-52. 1969.-The effects of phenobarbital pretreatment on the biliary excretion of four organic anions, indocyanine green (ICG), chlorothiazide (CTZ), phenol red, and probenecid, were investigated. Indocyanine green and CT2 are not metabolized in the rat, whereas phenol red and probenecid are excreted into the bile largely as glucuronides. Phenobarbital-pretreated male rats excreted more ICG and probenecid, but not more phenol red or CTZ, than controls. Females pretreated with phenobarbital excreted more CT2 and probenecid, but not greater amounts of ICG or phenol red. In the induced animals the glucuronides of phenol red and probenecid were not excreted in greater amounts than in control animals. Where increased biliary excretion of these compounds were observed, it was concluded to be due primarily to the increased bile flow in phenobarbital-treated animals.

Gender difference in the Oatp1-mediated tubular reabsorption of estradiol 17beta-D-glucuronide in rats.

The gender difference in the urinary excretion of estradiol-17beta-glucuronide (E(2)-17betaG) was examined in rats. The urinary clearance of E(2)-17betaG was >250 times lower in male than in female rats. No such major gender difference was observed in its biliary excretion or metabolism in kidney homogenate. Both plasma protein binding and inulin clearance were comparable in male and female rat...

Effect of ethynylestradiol on biliary excretion of bile acids, phosphatidylcolines, and cholesterol in the bile fistula rat.

The effects of ethynylestradiol on endogenous bile acids, their capacity to conjugate and excrete intravenously infused cholic acid, the concentrations of biliary cholesterol and lecithin, and the individual molecular species of phosphatidylcholine have been determined in male and female Sprague-Dawley rats. Endogenous biliary bile acids were analyzed by gas-liquid chromatography-mass spectrome...

Gender difference in the urinary excretion of organic anions in rats.

This study was aimed at clarifying the gender differences in the urinary excretion of organic anions and the gene expression of organic anion transporters in rats. The renal clearance with regard to the plasma concentration (CL(urine,p)) of taurocholate, dibromosulfophthalein (DBSP), and zenarestat, all substrates and/or inhibitors of organic anion transporting polypeptide 1 (Oatp1), was much h...

The effect of AT2 and Mas receptors antagonists on renal hemodynamic and excretory disorders induced by ischemia/reperfusion in male and female rats

Introduction: Renal ischemia-reperfusion (RIR) may disturb renin-angiotensin system components. In this study, the effects of Mas receptor (A779) and AT2 receptor (PD123319) antagonists were examined in RIR rats. Methods: Total 60 male and female Wistar rats were assigned into 10 groups (n=6 in each group), including sham-operated group, RIR groups treated with the vehicle, A779, PD123319, ...

Phenobarbital alters hepatic Mrp2 function by direct and indirect interactions.

Phenobarbital (PB) treatment impairs the biliary excretion of some organic anions. One mechanism may involve direct competition for biliary excretion by PB and/or a PB metabolite. Alternatively, PB may alter the expression and/or function of hepatic organic anion transport proteins. The role of multidrug resistance-associated protein 2 (Mrp2) in the biliary excretion of PB and metabolites was s...
